NUOVI FARMACI BIOTECNOLOGICI ATTIVI ATTRAVERSO LA MODULAZIONE DELL’ATTIVITÀ RECETTORIALE

Description The present project contains activities of both industrial research and industrial development, to develop and validate novel molecules acting as mimetics of neurotrophins and molecules inhibiting neo-angiogenesis. Recently, some of the Research Units of the present Project have set up: Line 1) novel low molecular weight non-peptidic compounds that possess metabolic activities in vitro superimposable to those of native NGF: such molecules selectively interact with TrkA receptor molecule, induce its autophosphorylation and activate downstream signalling pathways, thus modulating the function of key enzymes and proteins essential for cell survival (MAPK, ERK, Bcl-2, etc.): on this basis, they are essentially mimetics of the native neurotrophin. One of the major activities of NGF mimetics so far detected is the marked inhibition of the apoptotic process, consistent with the activities of NGF itself. This experimental line shall therefore be devoted to setting up pharmaceutical preparations for the following diseases: neurodegenerative disorders (such as Alzheimer disease or Parkinson disease); pressure or ischemic ulcers; neurotrophic corneal ulcers and glaucoma retinal damage; acute myocardial or cerebral ischemia; neoplastic disorders, particularly of the Central Nervous System. Line 2) drugs originating from synthetic peptides able to inhibit neoangiogenesis through the inhibition of endothelial cell migration. The family of peptides under investigation is composed of low molecular weight compounds, easily prepared through chemical synthesis and easily purified through low cost industrial procedures. As the main activity of such peptides is the blockade of new vessels growth (anti-angiogenic activity), their potential applications focus onto diseases taking advantage from inhibition of neoangiogenesis, such as the growth or metastatization of solid tumors, the neovascular proliferation of diabetic retinopathy or of senile macular degeneration, particularly in its wet form. Within the present project, such compounds will be studied for their use in diabetic retinopathy and senile macular degeneration. Both Project Lines include Phase I and Phase II clinical studies.
