Antigeni e adiuvanti per vaccini e immunoterapia

Description This project aims at reaching three main objectives: 1. Development of innovative, highly effective and safe vaccines against viral and bacterial infections, through the use of novel technologies for genome identification and analysis; 2. The set up of novel adjuvant molecules and the study of the activity mechanisms of molecules already in use or in development; 3. The development of new, safer and more effective viral vectors for the realisation of new vaccines against infective pathologies. This project develops a complete strategy that, by starting from the reverse vaccinology, leads to the rational design of innovative vaccines against selected pathogens. The high potentiality of this integrated approach will be applied to Streptococcus pneumoniae, Clostridium difficile, Escherichia coli and Human Cytomegalovirus (HCMV) in order to obtain vaccine candidates to be industrially tested. The identified antigens will be tested for their immunogenicity and biochemical stability in the presence of appropriate adjuvants (already known or in development) that will be studied and characterised in the second work package of the project. Animal immunisation experiments will then be necessary to properly evaluate the level of the immune response in relation to the administered adjuvant. Furthermore, this project will take advantage of a development strategy of new vaccines based on viral vectors (such as Adenovirus and MVA, VLPs) able to induce powerful cellular immune responses. The extremely innovative approach proposed in this project, based on a high level and cutting edge public-private partnership (predominantly from “Aree di Convergenza” such as Sicilia, Calabria and Campania), is a unique example of innovation of the process for identifying and obtaining new vaccines.
