Description This project aims to understand the molecular mechanisms of cancer pathology with the final scope to identify biomarkers as molecular targets of new anticancer substances and safety biomarkers as preclinical signals of drug induced vascular injury. Two fields of great interest for the human health system and, as consequence, for pharmacological research, are those related to neoplasie and to drug-induced side effects. In fact at epidemiological level neoplasie constitute the second cause of death after cardiovascular pathologies. The impact of such pathologies in the industrialized countries follows an exponential trend similar to the increase registered in the average survival index of population (aging) causing a significant growth in the pharmaceutical oncology market. In the pharmacological research field the efforts are aimed to obtain new biologics that more effectively hit the targets while diminishing the unpleasant side effect commonly associated to their use. In fact to date almost all the available drugs are characterized by a high therapeutic index that is counterbalanced by negative unavoidable side effects that include a wide range of pharmacological reactions, some of toxic nature, which can appear during the treatment. One of these negative side effects is represented by drug-induced vascular injury and, at present, new technologies such as genomics, proteomics and bioinformatics, represent a valid tool to characterize the molecular mechanisms linked to lesions. To resolve these issue the scientific research in general, and biopharmaceutical research in particular, aim to indentify new biomarkers and to develop innovative diagnostic and therapeutic tools. The present project focus on two objectives: the identification and validation of new biomarkers studying the molecular bases of oncological pathologies and the mode of action of antitumor compounds; understanding of those mechanisms responsible for some negative side effects related to the use of drugs like those associated with vascular damage. At present, biomedicine is in the midst of a technological revolution, driven by advances in genomics, proteomics, metabolomics and bioinformatics. Scientists strive to explore biomarkers and their possible role in the diagnosis, treatment and prognosis of tumor diseases. Researchers are trying to develop better screening and treatment options in the fight against the malignancy, as well as searching for proteins that may serve as biomarkers to detect and locate incipient cancer, serve as possible drug target, monitor response to therapy and predict the chance of recurrence after treatment has ended. Moreover, being biomarkers indicative of a biological change caused by the drug, they are an essential mean of pharmaceutical research that allows selecting, at an early stage of their development, the most successful molecules. Aim of this project will be the following areas of research: Identification of new molecular targets in the field of vascular biology; Research on new therapeutic targets to develop more efficacious and safe antitumor drugs; Implementation of databases of gene and protein expression profile as comparative and predictive tool for biomarkers identification and drug development.
